Allele-Specific Detection Methods for QoI Fungicide-Resistant <i>Erysiphe necator</i> in Vineyards

Grapevine powdery mildew (GPM), caused by the fungus Erysiphe necator, is a constant threat to worldwide production of grape berries, requiring repeated use fungicides for management. The frequent fungicide applications have resulted in resistance commonly used quinone outside inhibitor (QoI) and associated with single-nucleotide polymorphisms (SNPs) mitochondrial cytochrome b gene (cytb). In this study, we attempted detect most common SNP causing glycine alanine substitution at amino acid position 143 (i.e., G143A) cytb protein, track using allele-specific TaqMan probe digital-droplet PCR-based assays. Specificity sensitivity these assays showed that two could discriminate SNPs were effective on mixed samples. These diagnostic implemented survey E. necator samples collected from leaf air California Oregon grape-growing regions. Sequencing PCR amplicons phenotyping isolates also revealed accurately detected each allele (100% agreement), there was an absolute agreement between presence or absence G143A mutation QoIs sampled. results indicate developed tools will help growers make informed decisions about selections which, turn, facilitate GPM disease management improve systems.
